  • INFLUENCES OF ZINC ON CONCENTRATIONS OF CYCLIN D2 AND CYCLIN-DEPENDENT KINASE 4 OF HUMAN UMBILICAL CORD BLOOD-DRIVED MESENCHYMAL STEM CELLS

    Objective To explore effects of zinc on the contents of cycl in D2, cycl in-dependent kinase 4 (CDK4), and their DNA and total cellular protein in human umbil ical cord blood-drived mesenchymal stem cells (hUCBMSCs). Methods hUCBMSCs were isolated and cultured by density gradient centrifugation adherence method in vitro. At the serial subcultivation, the hUCBMSCs were randomly divided into 7 groups. In control group, hUCBMSCs were cultured with DMEM medium (containing 15%FBS). In treatment groups, hUCBMSCs were cultured with DMEM medium (containing 15%FBS plusZnSO4•7H2O). The final concentrations of zinc were 0.5, 1.5, 2.5, 3.5, 4.5, and 5.5 mg/L, respectively. The cellular surface antigens of CD29, CD34, CD44, and CD45 at the 3rd generation of hUCBMSCs were detected by flow cytometry. MTT assay was used to detect cell activity of the 3rd generation of hUCBMSCs. The optimum concentration of zinc was selected by the results of MTT as experimental group. The cell growth curves of experimental group and control group were drown by counting cell. The cell surface antigen, reproductive cycle, and DNA content were detected by flow cytometry motheds. The contents of cycl in D2 and CDK4 were detected by Western blot method. Results The positive expression rates of CD29 and CD44 were more than 70% in hUCBMSCs. The cell activity of 2.5 mg/L treatment group was superior to other treatment groups, as experimental group. At 7, 14, and 28 days, the contents of DNA, total cellular protein, cycl in D2, and CDK4 of hUCBMSCs were significantly higher in experimental group than those in control group (P lt; 0.01). The percentage of hUCBMSCs at S stage and prol iferation index in experimental group were also significantly higher than those in control group (P lt; 0.01). Conclusion Zinc (0.5-4.5 mg/L) has the promoting effect on the hUCBMSCs activity, and 2.5 mg/L is the optimal concentration. Zinc (2.5 mg/L) can accelerate the prol iferation and DNA reproduction of hUCBMSCs and increase the contents of cycl in D2 , CDK4, and cellular total protein.

  • OBSERVATION OF REPLICATIVE SENESCENCE OF RAT CHONDROCYTES IN VITRO

    Objective To observe the replicative senescence of rat articular chondrocyte cultured in vitro so as to provide reference for the succeeding experiment of using medicine interfere and reverse the cataplasia of tissue engineering cartilage or probing cataplasia mechanism.Methods Different generations(P1, P2, P3 and P4) of the chondrocytes were detected with the methods of histochemistry for β-galactosidase (β-gal), electronmicroscope for ultromicrostructure, immunocytochemistry for proliferating cell nuclear antigen (PCNA),alcian blue stain for content and structure of sulfatglycosaminoglycan (GAG) of extracellular matrix (ECM),reverse transcriptionpolymerase chain reaction (RTPCR) for content of collagen Ⅱ,flow cytometry for cell life cycle and proliferative index(PI) to observe senescence of chondrocytes.Results In the 4th passage,the chondrocytes emerging quantitively positive express of β-gal,cyto-architecture cataplasia such as caryoplasm ratio increasing and karyopycnosis emerging under electronmicroscope ,cell life cycle being detented on G1 phase(83.8%),while in P1, P2, P3 the content of G1 phase was 79.1%, 79.2%, 80.8% respectively. In the 4th passage, PI decreased(16.2%),while in P1, P2, P3, it was 20.9%, 20.8%, 19.2%. The positive percentage of PCNA,the content of GAG(long chain molecule) and the positive expression of collagen Ⅱ diminished,all detections above were significantly different (Plt;0.01) when compared the 4th passage with the preceding passages.Conclusion Chondrocytes show the onset of senescence in the 4th passage.

  • Effect of 8-bromum-cyclic adenosine monophosphate on the telomerase activity and dynamics in retinoblastoma cells

    Objective To investigate the effect of the 8-bromum-cyclic adenosine monophosphate (8-Br-cAMP) on the telomerase activity and changes of cell cycle in retinoblastoma (RB) cells. Methods The cultured RB cells were divided into the experimental group (8-Br-cAMP) and control group. After cultured for 24, 48 and 72 hours in vitro, the telomerase activity of RB cells was detected by polymerase chain reaction enzyme-linked immunosorbent assay (PCR-ELISA) and the changes of cell cycle were detected by flow-cytometry. Results The difference of telomerase activity was significant between the experimental groups and control group (Plt;0.01). There was a negative correlation between the A value of absorbance and the time in the experimental groups (r=-0.778 9, F=33.936, Plt;0.01). The changes of the cell cycle were that the percentages increased in G1 phase and decreased in S phases. Conclusion 8-Br-cAMP may weaken telomerase activity, affect the cell cycle, and inhibit the proliferation of RB cells. (Chin J Ocul Fundus Dis,2004,20:358-360)

  • THE EXPRESSION OF CELL CYCLE-REGULATED PROTEINS IN PANCREATIC ADENOCARCINOMA AND ITS SIGNIFICANCE

    Objective To know the abnormal expression of the cell cycle-regulated proteins in pancreatic adenocarcinoma and their effect on tumor cell growth. Methods The expression of p16, p21, Rb and p53 protein in 47 cases were investigated by immunohistochemistry with wet autoclave pretreatment for antigen retriaval. Furthermore, tumor growth index were assessed by a novel anti-ki-67 antibody (ki-s5). Results All the expression of p53, p16, p21 and Rb protein were the nuclear stainning. The positive rates of p53, p16, p21 and Rb protein were 55%, 53%, 74% and 98% respectively. There was negative correlation between of p16, p21 or Rb protein expression and ki-67 growth index. No relation of p53 protein stainning and the expression of p21 protein was found. Conclusion In pancreatic adenocarcinoma, the negative expression of p16 protein and p21 protein may play an important role in tumor cell growth, but tumor proliferation caused by abnormality of Rb protein is rare. The expression of p21 protein was not associated with the expression of p53 protein.

  • Relationship between cyclin dependent kinase 5 and apoptosis in the pathogenesis of retinitis pigmentosa in Royal College of Surgeon rats

    Objective To observe the expression of cyclin dependent kinase 5 (Cdk5) and p25 in the pathogenesis of retinitis pigmentosa (RP) in Royal College of Surgeon (RCS) rats and its relationships with apoptosis. To explore the mechanism of Cdk5 and p25 induced photoreceptor apoptosis in the pathogenesis of RP. Methods Retinas of RCS and RCS-rdy+ rats were obtained at the ages of postnatal day 17, 25, 35, 60. The retinal structure and thickness of outer nuclear layer were measured by optical microscopy. The expression of Cdk5, p25, cleave-caspase 3 in the retina was evaluated by immunohistochemistry. The protein expression of cleave-caspase 3 in the retina was determined by Western blot. The apoptosis of retinal cells was examined by terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL). The mean absorbance value of apoptotic cells was analyzed by SPSS 17.0 software. Results The retinal thickness of the RCS rats was significantly reduced in comparison to the RCS-rdy+ rats as the postnatal days progressed, particularly in the layer of rods and cones and the outer nuclear layer. The expression level of Cdk5, p25, cleave-caspase 3 of RCS rats increased from postnatal 17 days to postnatal 35 days, while decreased on postnatal 60 days; but there was no obvious change of above indexes in RCS-rdy+ rats. The protein expression of cleave-caspase 3 in the RCS rats was significantly increased with progression of postnatal days to postnatal 35; but there was no obvious similar change in RCS-rdy+ rats. The results of TUNEL showed that the apoptotic cells significantly increased in the outer nuclear layer of RCS rats from postnatal 17 days to postnatal 35 days, while decreased on postnatal 60 days; but there was no obvious change of above index in RCS-rdy+ rats. This study showed that there were significant correlations between the following variables: Cdk5 expression and p25 expression, Cdk5 expression and cleave-caspase 3 expression, Cdk5 expression and apoptotic cells, p25 expression and cleave-caspase 3 expression, p25 expression and apoptotic cells, cleave-caspase 3 expression and apoptotic cells. The partial correlation coefficients were 0.949, 0.808, 0.959, 0.887, 0.979, 0.852, respectively and the P value was 0.000. Conclusions The apoptotic cells significantly increases and the expression level of Cdk5, p25, cleave-caspase 3 of RCS rats increases from postnatal 17 days to postnatal 35 days. The tendency of apoptotic cells to increase is consistent with the change of Cdk5, p25, cleave-caspase 3 expression. The apoptosis of photoreceptor cells is related to increasing expression of Cdk5 and p25 in RCS rats. Cdk5 may be involved in the development of RP in RCS rats.

  • Effects on 5-Fluorouracil Induced Apoptosis and Cell Cycle Changes via Blocking Human Equilibrative Nucleoside Transporters in Pancreatic Cancer Cell Membrane

    ObjectiveTo investigate the effects of dipyridamole (DP), one of the human equilibrative nucleoside transporters (hENTs) blockers, on 5-fluorouracil (5-FU) induced cell apoptosis and cell cycle changes of the pancreatic cancer Panc-1 cell line. MethodsPancreatic cancer cell line Panc-1 was divided into hENTs blocked group and hENTs unblocked group. The hENTs blocked group was further divided into two subgroups according to the DP concentration, 5 μmol/L DP group and 10 μmol/L DP group. Each group was incubated in culture medium with or without 1.5×106 ng/L 5-FU for 24 h. Cell apoptosis and cell cycle were detected by flow cytometry. Results①The apoptosis results of each group: Incubated in culture medium with 1.5×106 ng/L 5-FU for 24 h, the apoptosis rates of 5 μmol/L DP group and 10 μmol/L DP group were higer than those of hENTs unblocked group (Plt;0.05), and which of 10 μmol/L DP group was higer than that of 5 μmol/L DP group (Plt;0.05). Incubated in culture medium without 5-FU for 24 h, there were no significant differences of the apoptosis rates among three groups (Pgt;0.05). ②The cell cycle results of each group: Incubated in culture medium with 1.5×106 ng/L 5-FU for 24 h, the percentages of S phase cells in the 5 μmol/L DP group and 10 μmol/L DP group were less than those of hENTs unblocked group (Plt;0.05). The percentage of S phase cell of 5 μmol/L DP group reduced to 87.09% and that of 10 μmol/L DP group reduced to 74.06% as compared with hENTs unblocked group. 5-FU had little influence on G2 phase (Pgt;0.05) except for the percentage of G2 phase cells in the 5 μmol/L DP group increased significantly (Plt;0.05) as compared with the hENTs unblocked group. Incubated in culture medium without 5-FU for 24 h, there were no significant differences of the cell cycles among three groups (Pgt;0.05). ConclusionsIn pancreatic cancer Panc-1 cell, DP could enhance the cytotoxicity of 5-FU by blocking hENTs. The enhanced cytotoxicity is related to elevation of 5-FU concentration in cells, and unrelated to DP itself.

  • EXPRESSION AND SIGNIFICANCE OF CYCLIN D1, CYCLINE AND P27 IN GASTRIC CARCINOMAS

    Objective To study the expression and its significance of cyclin D1,cyclin E and p27 protein in gastric carcinoma. Methods The expression of cyclin D1,cyclin E and p27 protein in 60 cases of gastric carcinoma were detected by immunostaining method. Results The positive rates of cyclin D1,cyclin E and p27 protein expression in gastric carcinoma were 41.7%(25/60),38.3%(23/60)and 36.7%(22/60),respectively.The expression of cyclin D1 was significantly correlated with Lauren type of tumor,the expression of cyclin E was significantly correlated with tumor differentiation,infiltration and stage,while the expression of p27 protein was significantly correlated with tumor infiltration,lymph nodes metastasis and stage. Further more, the expression of cyclin E was inversely correlated with the expression of p27. Conclusion Detection of cyclin D1,cyclin E and p27 expression in gastric carcinoma may be helpful in determining tumor progression.

    Objective To investigate cell cycle as a new tool to evaluate the biocompatibility of biomaterials.Methods The cell cycle and the expression of related genes were analyzed by the methods of immunocytochemistry, protein blotting, RT PCR and flow cytometry. Results The physical properteis, chemical properties and topological properities of biomaterials could not only influence cell cycle of the cells attached onto biomaterials but also affect the expression of related genes of target cells. Conclusion As an important extension of routine proliferation epxeriments, the study of cell cycle control will be great help for us to to study the cell group as an organic society. It revealed the balance between cell proliferation, cell differentiation and apotosis. It is suggested that the study of cell cycle control will play a key role in the research of tissue engineering.

  • Effect of Multiple Thermotherapy on Proliferation Cycle and bcl-2 mRNA of HepG2 Cell

    【Abstract】ObjectiveTo investigate the proliferation rate of HepG2 cell after multiple thermotherapy and the possible reasons related to it. MethodsAfter HepG2 cell were treaded by ten repeated cycles of heat exposure at 43 ℃ for 80 minutes twice a day, the doubling time of cell was analyzed, and the cell cycle, bcl2 mRNA and bax mRNA were detected. ResultsThe proliferation rate of HepG2 cell which treated with heat speeded up, the percentage of G2 and S in cell cycle increased, the expression of bcl2 mRNA strengthened and the rate of bcl2/bax increased. ConclusionThe speeded proliferation of HepG2 cell after multiple thermotherapy is related to its high percentage of DNA duplicated and dividing cell, strengthened expression of bcl2 mRNA and increased rate of bcl2/bax.

  • FUNCTION OF MicroRNA-199b-5p IN EWING'S SARCOMA CELL LINES

    ObjectiveTo investigate the function and possible mechanism of microRNA-199b-5p (miR-199b-5p) in Ewing's sarcoma cell lines so as to provide theoretical basis for biological treatment in the future. MethodsA673 cells and TC252 cells were adopted as Ewing's sarcoma cell lines in vitro. miR-199b-5p oligonucleotide fragments (mimic) and scramble control (mimic control) were used to transfect TC252 cells and A673 cells, respectively. The expression of miR-199b-5p was measured in 2 cell lines by real-time quantitative-PCR and compared with that in mesenchymal stem cells (MSCs). The cell proliferation was examined by cell counting kit 8. The cell cycle and apoptosis were detected by flow cytometry. The possible targets of miR-199b-5p were determined by luciferase assays. The protein expressions of possible targets were measured by Western blot. ResultsThe expression of miR-199b-5p in control group was significantly down-regulated in A673 cells and TC252 cells when compared with that in MSCs (P<0.05); and the expression of miR-199b-5p in experimental group was significantly up-regulated when compared with that in control group (P<0.05). G1 phase cells increased obviously and S phase cells decreased significantly compared with corresponding cells in control group (P<0.05); but no significant difference was found in G2/M phase cells between 2 groups (P>0.05). The apoptosis rate increased significantly in experimental group when compared with that in control group (P<0.05). The possible targets of miR-199b-5p were cyclin-L1 (CCNL1) and c-Kit by luciferase assays. Western blot results showed that the CCNL1 and c-Kit protein expression levels in experimental group were significantly lower than those in control group (P<0.05). ConclusionmiR-199b-5p can suppress the cell proliferation, block the cell cycle, and promote the cell apoptosis, so miR-199b-5p inhibits Ewing's sarcoma cell lines by targeting CCNL1 and c-Kit.
