  • ANTISENSE RB GENE STIMULATES GROWTH OF HUMAN NORMAL CELLS

    Retinoblastoma susceptibility gene(Rb gene)is a tumor suppressor,which often deleted,mutated or inactivated in many malignant tumors.We construct an antisense Rb gene expression vector DOLRBAS and transfected it into human embryonic lung fibroblast(HEL cells)by electroporation technique.With the transient expression of antisense Rb gene the Rb protein synthesis is reduced and the growth rate of HEL cells in increased,but no coloney of HEL cells formed in soft agar.This result indicates that Rb gene can block cell division of HEL cells,but inactivation of Rb gene only is not sufficient for malignancy transformation of HEl cells.   • MOLECULAR GENETIC STUDY OF AUTOSOMAL DOMINANT CONGEITAL STATIONARY NIGHT BLINDNESS-ANALYSIS OF THE RHODOPSIN GENE BY PCR

    One of the major clinical characteristics of congenital stationary night blindness(CSNB)is dysfunction of rod photoreceptors of the retina.Rhodopsin,the photosensitive pigment of the rods,is essential for maintaining the normal function of rod photoreceptors.It is resonable to hypothesize that mutations or deletions of rhodopsin gene may be involved in the molecular defect of CSNB.To test this hypothesis,we are searching for rhodopsin gene mutations in patients with autosomal dominant CSNB.In this study,DNA fragments containing the coding sequences in exon 5 of rhodopsin gene were amplified by polymerase chain reaction(PCR)in 15 patients and 5 unaffected members from a large family with autosomal dominant CSNB.RFLP analysis of these DNA fragments demonstrated that in comparison with a control group of 12 normal persons,there is no obvious deletion in exon 5 of rhodopsin gene,and that mutations or deletions do not exist in codon 314,codon 347,and the third base of codon 313 as well as the first base of codon 348 of the rhodopsin gene in these CSNB patients,which suggest the molecular pathogenesis of autosomal dominant CSNB not involve mutations or deletions of these codons of the rhodopsin gene.   • Experimental research of effect of Rb gene on cell cycle of retinoblastoma transplantation tumor

    Objective To investigate the effect of exogenous Rb gene on the cell cycle of vitreous retinoblastoma (RB) transplantation tumor in nude mouse. Methods Based on establishing vitreous RB transplantation tumor in nude mouse,constructing retrovirus vector of Rb gene PBabe-Rb and transfecing it into the RB transplantation model by liposome Dosper,the change of cell cycle of the RB transplantation tumor by flow cytometry(FCM)was analysed. Results FCM showed that the cells of G1phase of the treated eyes were obviously more than the control eyes with the value of DNA index(DI)and S phase fraction(SPF) decreased by the Rb gene expression. Conclusion The exogenous PBabe-Rb gene can partially suppress the progress of the cell cycle of RB transplantation tumor in vivo. (Chin J Ocul Fundus Dis,2000,16:1-70)
