ObjectiveTo investigate the potential causal relationship between four types of reproductive behaviors and rheumatoid arthritis (RA), with the goal of establishing a theoretical foundation for clinical prevention and treatment strategies. MethodsPooled gene-wide association study (GWAS) data were obtained from large publicly searchable databases. Four characteristics like menarche, menopause, the age of first pregnancy and the age of last pregnancy, which related to reproductive behavior were selected as the exposure factors and RA as the outcome factors. Single nucleotide polymorphisms (SNPs), which were strongly correlated with the phenotype of the exposure factors, were screened as the instrumental variables, and two-sample Mendelian randomization analyses were used to assess the potential causal relationship between the exposure and the disease. Results① The Mendelian randomization analysis utilizing the inverse variance weighted method on two distinct samples revealed a significant negative correlation between the age of first pregnancy and last pregnancy with the risk of RA (OR=0.91, 95%CI 0.85 to 0.98, P=0.011; OR=0.54, 95%CI 0.31 to 0.93, P=0.026). Conversely, no causal relationship was observed between menarche and menopause with RA. Sensitivity analysis confirmed the robustness of the causal relationship, while MR Egger intercept analysis did not identify any potential horizontal pleiotropy (Page of first gestation -RA=0.169, Page of last gestation -RA=0.283). ② Reverse Mendelian randomization analysis revealed a significant positive causal association between RA and the age of first pregnancy, while no causal relationship was observed with the age of last pregnancy (OR=1.07, 95%CI 1.02 to 1.11, P=0.001). ③ Multivariate Mendelian randomization analysis demonstrated that both the age of first pregnancy and last pregnancy in women were inversely associated with the risk of RA (OR=0.88, 95%CI 0.80 to 0.97, P=0.010; OR=0.68, 95%CI 0.48 to 0.97, P=0.033). ④ There existed a negative correlation between the age of pregnancy in women and the risk of developing RA, suggesting a potential protective effect. ConclusionPregnancy age may have a negative causal relationship with the risk of RA, while menarche and menopause have no causal relationship with RA.
ObjectiveTo understand the clinical value of centromere proteins (CENPs) in hepatocellular carcinoma and their influence on the malignant biological behavior of hepatocellular carcinoma, and to provide theoretical references for related research in this field. MethodDomestic and international databases were searched for relevant literatures on the study of CENPs in hepatocellular carcinoma in recent years to be analyzed and reviewed. ResultsA total of 11 CENPs closely related to hepatocellular carcinoma had been summarized, which were differentially expressed in hepatocellular carcinoma and correlated with prognosis. CENPs might regulate various malignant biological behaviors such as hepatocellular carcinoma proliferation, metastasis, apoptosis, and resistance to radiotherapy and thus participate in the development of hepatocellular carcinoma via multiple mechanisms. The roles and mechanisms of some CENPs in hepatocellular carcinoma remained unclear. ConclusionsCENPs play an essential role in the diagnosis, treatment, and prognosis of hepatocellular carcinoma. They are involved in multiple malignant biological behaviors of hepatocellular carcinoma and are expected to be potential therapeutic targets for hepatocellular carcinoma. However, their roles and mechanisms in hepatocellular carcinoma remain to be investigated.
Objective To study the biological behavior of osteoblast and vascular endothelial cell culture. Methods The osteoblasts and vascular endothelial cells were obtained from calvarial bone and renal cortox of 2-week rabbits respectively. The experiment were divided into group A (osteoblasts), group B (vascular endothelial cells) and group C(co-cultured osteoblasts and vascular endothelial cells). The cells were identified with cytoimmunochemical staining. The cellular biological behavior and compatibilitywere observed under inverted phase contrast microscope and with histological staining. The cells viability and alkaline phosphatase(ALP) activity were measured. Results The cytoimmunochemical staining showed that the cultured cells were osteoblasts and vascular endothelial cells .The cellular compatibility of osteoblasts and vascular endothelial cells was good. The ALP activity was higher in group C than in group A and group B(P<0.01), and it was higher in group A than in group B(P<0.05). In group C, the cellproliferation were increased slowly early, but fast later. Conclusion Thecellular compatibility of osteoblasts and vascular endothelial cells were good. The vascular endothelial cells can significantly increased the osteoblast viability and ALP activity, and the combined cultured cells have greater proliferation ability.
ObjectiveTo explore the correlation of serum levels of 5-hydroxytryptamine (5-HT) and 5-HT2A receptor gene T102C polymorphism with suicidal behavior by poison in non-depressive patients, in order to diagnose and intervene the suicidal behavior of patients as early as possible. MethodsSixty-two non-depressive patients with the behavior of suicide by poison treated between January 2013 and June 2014 were selected as patient group, and other 66 healthy persons were selected as control group. Peripheral blood in the two groups of patients were collected to test the serum levels of 5-HT with enzyme linked immunosorbent assay, and the 5-HT2A receptor gene T102C polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism. The data were analyzed by SPSS 19.0 statistically. ResultsThe serum level of 5-HT in the patient group was significantly lower than that in the control group (P<0.001), and the frequency of T102C genotype in the patient group was higher than the control group (χ2=5.533, P=0.013). The distribution of genotype was different, and homozygous mutations of CC in the patient group were higher than the control group (χ2=5.648, P=0.017). ConclusionThe serum levels of 5-HT and 5-HT2A receptor gene T102C polymorphism, and the frequency of T102C genotype may be related to suicidal behavior by poison in non-depressive patients.
The material properties and volume proportion of the fibers as well as the cross-sectional area proportion of nucleus pulposus vary greatly in different studies. The effect of these factors on the mechanical behavior of intervertebral discs (IVDs) are uncertain. The IVDs finite element models with different parameters were created to investigate the pressure, height, rotation, stress, and strain of the IVDs under loads: pure compression, rotation after compression or axial moment after compression. The results showed that the material properties of fibers had great impact on the mechanical behavior of IVDs, especially on the rotation angle. When the fiber volume ratio was small, its changes had a significant impact on the rotation angle of the IVDs. The area proportions of nucleus pulposus had relatively little effect on the mechanical behavior of IVDs. The IVDs rotation should be observed when validating the model. By adjusting the elastic modulus or volume ratio of fibers within a reasonable range, a model that could simulate the mechanical behavior of normal IVDs could be obtained. It was reasonable to make the area proportion of nucleus pulposus within 25%–50% for the IVDs finite element model. This study provides guidance and reference for finite element modeling of the IVDs and the investigation of the IVDs degeneration mechanism.
Aiming at the limitations of clinical diagnosis of Parkinson’s disease (PD) with rapid eye movement sleep behavior disorder (RBD), in order to improve the accuracy of diagnosis, an intelligent-aided diagnosis method based on few-channel electroencephalogram (EEG) and time-frequency deep network is proposed for PD with RBD. Firstly, in order to improve the speed of the operation and robustness of the algorithm, the 6-channel scalp EEG of each subject were segmented with the same time-window. Secondly, the model of time-frequency deep network was constructed and trained with time-window EEG data to obtain the segmentation-based classification result. Finally, the output of time-frequency deep network was postprocessed to obtain the subject-based diagnosis result. Polysomnography (PSG) of 60 patients, including 30 idiopathic PD and 30 PD with RBD, were collected by Nanjing Brain Hospital Affiliated to Nanjing Medical University and the doctor’s detection results of PSG were taken as the gold standard in our study. The accuracy of the segmentation-based classification was 0.902 4 in the validation set. The accuracy of the subject-based classification was 0.933 3 in the test set. Compared with the RBD screening questionnaire (RBDSQ), the novel approach has clinical application value.
Carcinoembryonic antigen (CEA)was measured with ABC immunohistochemistry method in fourty-one gastric cancer tissues and sixty-six tissue from normal stomach and gastric benign lesions. The study revealed that the reactive signals in the former were ber than those in the latter. Simultaneously, CEA localized mainly in the cytoplasm or stroma in the cancerous tissue, but in normal gastric tissue or benign gastric lession, CEA distributed mainly in the margin of gland with gastric depression or membranous type. The result also revealed that the distribution patterns of ECA were linked with the cell growth types and infiltrating of gastric cancer. The authors consider that the expression state of CEA in gastric cancer is correlated with its biological behavior, and distribution patterns of CEA are more clinically significant than reactive intensities in the tissue. Patients have different prognosis with different CEA distribution patterns in tissue though their pathological types and TNM stages are the same.
Cognitive reappraisal is an important strategy for emotion regulation. Studies show that even healthy people may not be able to implement this strategy successfully, but the underlying neural mechanism behind the behavioral observation of success or failure of reappraisal is unclear. In this paper, 28 healthy college students participated in an experiment of emotional regulation with the cognitive reappraisal strategy. They were asked to complete the cognitive psychological questionnaires before the experiment. Their behavioral scores and scalp electroencephalogram (EEG) signals were collected simultaneously during the experiment. We divided all the subjects into two groups, according to the statistical test of valence scores. Then we analyzed their questionnaires, early event-related potential (ERP) components N200, P200, and late positive potential (LPP), and calculated the correlation between the valence score and the amplitude of LPP. The results showed that, in both groups, compared with negative-watching, the reappraisal induced larger N200 and P200 components and there were two modulation patterns (“increase” and “decrease”) of the reappraisal effect on the amplitude of early LPP (300−1 000 ms after stimulus onset). Moreover, correlation analysis showed that significant positive correlation between two differences in the successful group, i.e., the greater difference in the valence scoresin between reappraisal and negative-watching, the greater difference in the amplitude of early LPP between reappraisal and negative-watching; but no such effect was found in the failure group. These results indicated that, whether reappraisal was successful or not, no significant effect on early ERP components was found; and there were different patterns of the reappraisal effect on early LPP. The difference between successful and failure groups was mainly reflected in early LPP, that is, the EEG characteristics and behavioral scores of successful group were significantly positively correlated. Furthermore, the small sample analysis showed that this correlation only existed in the pattern of "increase". In the future, more research of this modulation mode is necessary in order to find more stable EEG characteristics under successful cognitive reappraisal in emotion regulation.
